Functional analysis of the XPF-like Zip2 protein and its partners in the resolution of meiotic recombination intermediates

Projet de stage :

Crossover formation upon repair of the meiotic DNA double-strand breaks is essential for the faithful segregation of homologs at Meiosis I. Crossovers are promoted by a group of proteins collectively termed « ZMM », whose molecular functions are poorly characterized. Using a proteomic approach, we have recently identified protein interactions between members of this family of proteins. The aim of the study will be to further characterize the protein interactions partners of Zip2, one of the ZMM proteins, and the functional domains involved. For this, the student will use two-hybrid and co-immunoprecipitation studies, as well as chromatin immunoprecipitation to investigate the binding sites of the protein along the genome. Complementary experiments will be to study the meiotic recombination phenotype of the inactivation of these proteins.
