Titre du projet de stage : The novel mechanisms of metabolic control by the class 3 PI3K

Projet de stage The proposed M2 project is a part of on-going research program in our laboratory that aims to discover how the class 3 PI3K controls metabolism in the cell. The best studied role of the class 3 PI3K is its function as a lipid kinase. Its activity is a major source of phosphoinosytol-3 phosphate in the cell which is an essential lipid messenger for endocytosis, autophagy and lysosomal function. The works of our team demonstrated a key role of the class 3 PI3K in a whole body metabolic homeostasis (Nemazanyy 2013, Nemazanyy 2015). In our recently submitted publication, we report an unappreciated requirement of the class 3 PI3K for mitochondrial function. Yet, the mechanisms of how the class 3 PI3K controls different metabolic processes are not known. We are looking for an enthusiastic, scientifically driven M2 trainee with a strong interest in signalling and metabolism to advance further the project that aims to discover how activity and subcellular localization of the class 3 PI3K impacts cellular metabolism. You will be integrated in our team to : (1) perform the microscopy and biochemical analyses that will uncover the subcellular localization of the class 3 PI3K (2) conduct the studies that will discover how sub-cellular localization of the class 3 PI3K affect cellular metabolism You will be trained in a highly dynamic scientifically stimulating French-English working language environment hand in hand with fellows from all over the world. The host laboratory is a part of a newly formed multidisciplinary international research centre in molecular medicine – the Institute Necker Enfants Malades (INEM) which emerged on a crossroad of fundamental, translational and clinical research. You will participate to the regular internal and external seminars held in our Institute and will have an opportunity to communicate with researchers of different levels and diverse research fields.

Techniques mises en œuvre par le stagiaire : You will work on the independent (though guided) research program under a mentorship of Dr. Ganna Panasyuk. You will master different wet lab molecular biology techniques/practices including cloning, generation of point mutants, RNA/protein extraction, RT-qPCR, immunoblot, immunohistochemistry, fluorescent microscopy, metabolomics approaches using masspectrometry and SeaHorse measurements, work with lentiviral and adenoviral vectors, work with mouse models and in vitro models (primary hepatocytes, mouse embryonic fibroblasts).
